Biological Background: hnRNP E2/PCBP2 Function and Localization
- Poly(rC)-binding protein 2 (PCBP2), also known as alpha-CP2 and hnRNP E2, is a single-stranded nucleic acid binding protein that preferentially binds oligo dC and poly(rC), with additional affinity for poly(rU).
Related references: PMID:12414943, PMID:7607214 - PCBP2 negatively regulates antiviral innate immunity: it functions as an adapter between MAVS and ITCH E3 ubiquitin ligase, leading to MAVS ubiquitination and degradation; it also inhibits the cGAS-STING pathway by interacting with CGAS to prevent liquid droplet formation.
Related references: PMID:19881509, PMID:35322803 - Together with PCBP1, PCBP2 is required for erythropoiesis, potentially through regulation of mRNA splicing.
- During poliovirus infection, PCBP2 binds the viral internal ribosome entry site (IRES) and stimulates IRES-mediated translation; it also plays a role in initiation of viral RNA replication in concert with the viral protein 3CD.
Related references: PMID:12414943, PMID:24371074 - PCBP2 localizes to both the nucleus and cytoplasm, consistent with its roles in RNA processing and cytoplasmic signaling.
- According to UniProt annotations, PCBP2 undergoes phosphorylation, ubiquitination, and alternative splicing, and contains multiple heterogeneous nuclear ribonucleoprotein K-homology (KH) domains typical of RNA-binding proteins.
- PCBP2 is ubiquitously expressed across all tissues, reflecting its fundamental roles in RNA metabolism.
Experimental Guidance and Technical Tips
- For Western blotting, an expected band is observed at approximately 39 kDa. Validate the antibody using positive control lysates from human, mouse, or rat cell lines.
- In immunoprecipitation assays, confirm antibody performance in your specific cellular context, considering the nuclear/cytoplasmic localization of PCBP2.
- For ELISA applications, consider using the synthetic peptide immunogen (amino acids 150–250) as a coating antigen to optimize detection sensitivity; cross-reactivity to orthologues should be verified in your assay format.
