Biological Background: hnRNP E1/PCBP1 Function and Localization

  • hnRNP E1/PCBP1 (Poly(rC)-binding protein 1) is a member of the heterogeneous nuclear ribonucleoprotein family, also known as Alpha-CP1, hnRNP E1, and nucleic acid-binding protein SUB2.3.
  • Functions as a single-stranded nucleic acid binding protein with high affinity for oligo dC sequences, playing critical roles in mRNA stabilization and translational regulation. Related references: PMID:15731341 PMID:7556077 PMID:7607214
  • Collaborates with its paralog PCBP2 to ensure proper erythropoiesis, likely through modulation of mRNA splicing events.
  • In the context of poliovirus infection, facilitates initiation of viral RNA replication in conjunction with the viral 3CD protein.
  • Subcellularly localizes to both the nucleus and cytoplasm, consistent with its roles in RNA metabolism and transport.
  • Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation, which may regulate its activity and interactions.
  • Tissue expression is abundant in skeletal muscle, thymus, and peripheral blood leukocytes, with lower levels in prostate, spleen, testis, ovary, small intestine, heart, liver, adrenal, and thyroid glands.

Experimental Guidance and Technical Tips

  • The antibody is raised against a synthetic peptide corresponding to amino acids 100-200 of human hnRNP E1/PCBP1; it is predicted to detect the endogenous 37kDa protein in appropriate samples.
  • For Western blotting, consider initial dilutions of 1:500–1:2000 and validate using known positive lysates from skeletal muscle, thymus, or leukocyte lines.
  • In immunohistochemistry (IHC-P), antigen retrieval may be required; optimization using positive control sections of skeletal muscle or thymus is recommended.
  • For immunofluorescence (IF/ICC), permeabilization steps are necessary to access nuclear and cytoplasmic compartments; counterstain with nuclear marker to confirm localization.
  • ELISA applications may be used for titering or antigen capture; however, optimal antigen coating and detection conditions should be determined empirically.
