Biological Background: hnRNP A1 Function and Localization

  • Heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1), also known as helix-destabilizing protein or single-strand RNA-binding protein, is an RNA-binding protein encoded by the HNRNPA1 gene (Gene ID: 3178).
  • Involved in packaging pre-mRNA into hnRNP particles, transporting poly(A) mRNA from nucleus to cytoplasm, and modulating splice site selection. Related references: PMID:17371836
  • Plays a role in splicing of pyruvate kinase PKM by repressing sequences flanking PKM exon 9, inhibiting exon 9 inclusion, leading to exon 10 inclusion and production of the PKM M2 isoform. Related references: PMID:20010808
  • Binds to the IRES and inhibits translation of apoptosis protease activating factor APAF1. Related references: PMID:31498791
  • May bind to specific miRNA hairpins. Related references: PMID:28431233
  • Localizes to both nucleus and cytoplasm, participating in nucleocytoplasmic shuttling of mRNA.
  • Undergoes acetylation, methylation, phosphorylation, and ubiquitin conjugation; disease variants are associated with amyotrophic lateral sclerosis (ALS19, ALS20) and neurodegeneration.
  • May play a role in HCV RNA replication; cleavage by Enterovirus 71 protease 3C leads to increased APAF1 translation and apoptosis.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1‑140) of human hnRNP A1; the antibody may recognize full-length and N-terminal fragments. KO validation supports specificity in western blotting.
  • For western blotting, use whole-cell or tissue lysates from human, mouse, or rat. Include appropriate controls and validate detection of the ~39 kDa band.
  • For immunofluorescence/ICC, expect nuclear and cytoplasmic staining. Optimize fixation and permeabilization conditions for the relevant cell line.
  • For ELISA, test against recombinant hnRNP A1 protein or cell lysates; confirm linearity and detection limit in the experimental system.
