  • HNF3β/FOXA2, also known as Forkhead box protein A2 or TCF-3B, is a transcription factor encoded by the FOXA2 gene in humans.
  • Acts as a pioneer transcription factor by binding nucleosomal core histones, displacing linker histones, and opening compacted chromatin at target enhancers/promoters to facilitate gene regulation (By similarity).
  • Essential for embryonic development, including notochord formation and development of endoderm-derived organs such as the liver, pancreas, and lungs; functions partially redundantly with FOXA1.
  • Regulates expression of liver-specific genes like AFP, albumin, tyrosine aminotransferase, and PEPCK by interacting with their cis-regulatory elements.
  • Involved in glucose homeostasis by controlling pancreatic β-cell gene expression and modulates fat metabolism; binds fibrinogen beta promoter and mediates IL6-induced transcriptional activation.
  • Localizes predominantly to the nucleus but can also be found in the cytoplasm.
  • Post-translationally modified by phosphorylation; its DNA-binding domain recognizes the consensus sequence 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3'.
