Biological Background: Hmox1 Function and Localization

  • Heme oxygenase 1 (Hmox1), also designated HO-1 or P32 protein, is a microsomal enzyme that catalyzes the rate-limiting step in heme degradation.
  • It cleaves heme at the alpha-methene bridge to produce equimolar amounts of biliverdin IXalpha, carbon monoxide (CO), and ferrous iron.
  • Hmox1 confers cytoprotection by eliminating pro-oxidant free heme, thereby preventing heme-induced apoptosis.
  • The protein is anchored to the endoplasmic reticulum membrane via a single C-terminal transmembrane helix.
  • Classified as an oxidoreductase with metal-binding capacities for heme and iron, Hmox1 is also a phosphoprotein.
  • Belongs to the heme oxygenase family and plays a critical role in cellular iron homeostasis and antioxidant defense.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1-266 of mouse Hmox1 (NP_034572.1), covering the majority of the protein; thus the antibody is expected to recognize both denatured and native epitopes.
  • For western blot (WB), expect a band at approximately 33 kDa. Include positive controls such as mouse spleen, liver, or kidney lysates, where Hmox1 is highly inducible.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), validate staining specificity using tissues from untreated and hemin- or stress-induced animals.
  • For ELISA, consider using the recombinant immunogen as a reference standard to assess antibody binding and specificity.
  • Cross-reactivity has been confirmed with rat samples; verify performance in other species if needed.
