Biological Background: HLTF Function and Localization
-
HLTF (Helicase-like transcription factor, also known as SMARCA3, RNF80, HIP116, SNF2L3) is a dual-function enzyme with DNA-dependent ATPase activity and E3 ubiquitin-protein ligase activity, involved in chromatin regulation and DNA damage tolerance (DDT).

It catalyzes 'Lys-63'-linked polyubiquitination of monoubiquitinated PCNA at 'Lys-164', promoting error-free postreplication repair via template switching.
Related references: PMID:18316726, PMID:18719106 -
HLTF binds histone H3 trimethylated at 'Lys-9' (H3K9me3) and mediates histone H3 'Lys-23' polyubiquitination (H3K23ub), creating a docking site for DNMT1 to ensure replication-coupled DNA methylation maintenance.

As a chromatin remodeling factor, HLTF regulates transcription and exhibits ATP-dependent double-stranded DNA translocase activity without classical helicase function; it can mediate replication fork reversal to suppress DNA synthesis and maintain genomic stability.
Related references: PMID:10391891, PMID:1994885, PMID:9126292 -
HLTF cooperates with MSH2 to resolve G-quadruplex (G4) DNA structures, limiting replication stress and G4 accumulation throughout the cell cycle.
Related references: PMID:39142279 -
It contributes to nucleotide excision repair by evicting lesion-containing oligonucleotides via its HIRAN and ATPase domains, and can displace single-stranded DNA from triplex structures.
Related references: PMID:26051180, PMID:31960921 -
HLTF also has protein clearing activity at stalled replication forks, facilitating restart of DNA replication.
Related references: PMID:21795603 -
HLTF localizes to the nucleus and chromosomes, and is broadly expressed in tissues including brain, heart, kidney, liver, lung, pancreas, placenta, and skeletal muscle.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 726–1009 of human HLTF, covering a C‑terminal region that includes the RING finger domain; this design may help generate antibodies recognizing full‑length HLTF and its key functional epitopes.
- For Western blot, the expected molecular weight of HLTF is approximately 114 kDa; validate the antibody in relevant human cell lines, as cross‑reactivity with other species has not been tested.
- This antibody is intended for ELISA and WB applications; consider optimizing antibody dilutions and assay conditions for your specific sample matrix and experimental system.
- Shipment is on ice bags; upon receipt, store the antibody at the recommended temperature (–20°C or –80°C) to maintain stability.
