Biological Background: HLA-B Function and Localization
- HLA-B, also known as human leukocyte antigen B, is a classical major histocompatibility complex class I (MHC I) heavy chain that assembles with β2-microglobulin (B2M) to form a functional antigen-presenting complex on the cell surface.
- It plays a central role in adaptive immunity by presenting primarily viral and tumor-derived peptides of 8-13 amino acids to αβ T cell receptors (TCR) on CD8+ T cells, triggering antigen-specific cytotoxic responses. - HLA-B is highly polymorphic with over 9000 alleles; different alleles exhibit distinct peptide-binding motifs. For example, allele B07:02 preferentially binds peptides with Pro at position 2 and a C-terminal Leu, while B27:05 requires an Arg at position 2.
