Biological Background: Histone H1.2 Function and Localization
- Protein identity: Histone H1.2 (also known as H1c, H1d, H1s-1) is a member of the histone H1 family, encoded by the H1-2 gene (HIST1H1C) in humans.
- Chromatin compaction: Histone H1.2 binds linker DNA to form the chromatin fiber and is essential for condensation of nucleosome chains into higher-order structures; its loss leads to global chromatin decompaction. Related references: PMID:26581166, PMID:38530350
- Histone reader activity: Specifically recognizes and binds histone H3 trimethylated at lysine 27 (H3K27me3). Related references: PMID:26581166
- Role in Polycomb repression: Promotes H3K27me3 methylation by the PRC2 complex, facilitating epigenetic maintenance. Related references: PMID:37429872, PMID:40516528
- Splicing regulation: Together with histone H1.3, regulates alternative splicing, particularly exon skipping and intron retention, by modulating RNA polymerase II elongation. Related references: PMID:37922872
- Gene transcription modulation: Regulates individual gene transcription through chromatin remodeling, nucleosome spacing, and DNA methylation. Related references: PMID:38530350
- Subcellular localization: Predominantly nuclear, enriched in the nucleolus and chromosomes.
- Post-translational modifications: Subject to acetylation, methylation, phosphorylation, citrullination, ADP-ribosylation, and hydroxylation, which modulate chromatin-binding affinity and function.
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 100-200 of human HIST1H1C, a region within the globular domain. Verify specificity in your experimental system, as this domain is conserved among histone H1 variants.
- For Western blotting, expect a band at approximately 21 kDa. Optimize blocking and washing steps to minimize nonspecific binding.
- In IHC-P and IF/ICC, nuclear staining should be observed. Use appropriate antigen retrieval and fixation protocols.
- For ELISA, this antibody may be used for detection or capture; validate compatibility when developing a matched pair.
- Cross-reactivity with mouse and rat is reported; confirm species specificity with control lysates and recombinant proteins.
