Biological Background: Histamine H2 Receptor Function and Localization
- Histamine H2 receptor (HRH2, also known as H2R, HH2R, or gastric receptor I) is a G protein-coupled receptor encoded by the HRH2 gene in humans and exists in several alternatively spliced isoforms.
- Upon binding histamine, HRH2 predominantly couples to G(s) alpha proteins, activating adenylate cyclase and increasing intracellular cAMP levels, which stimulates gastric parietal cells to secrete hydrochloric acid (PubMed:38647423, PubMed:39333117). Related references: PMID:38647423 PMID:39333117
- HRH2 is also expressed in the heart and central nervous system, where it may contribute to cardiac stimulation and modulate neurotransmitter release.
- The receptor localizes to the cell membrane, consistent with its role as a transmembrane signaling protein.
- Key post-translational modifications include glycosylation, palmitoylation, and the formation of disulfide bonds, as annotated in UniProt keywords.
- The molecular weight of the mature protein is approximately 40 kDa.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide corresponding to amino acids 150-250 of human HRH2. Consider sequence alignment with mouse and rat HRH2 to confirm epitope conservation across species when planning cross-species experiments.
- This antibody has been listed for Western blot (WB) and ELISA. For WB, a band at ~40 kDa is anticipated. Validate the antibody in your sample system, as detection efficiency can vary with sample preparation and expression levels.
- When using ELISA, titrate the antibody and include appropriate controls to ensure specificity.
