Biological Background: HIPK2 Function and Localization

  • HIPK2 is a serine/threonine kinase that phosphorylates key regulators such as p53/TP53, CREB1, and CTNNB1, inhibiting cell growth and promoting apoptosis.
  • Acts as a transcriptional corepressor for factors like SMAD1 and POU4F1, and modulates Wnt/beta-catenin signaling by promoting proteasomal degradation of CTNNB1 and MYB.
  • In response to DNA damage, HIPK2 phosphorylates DAZAP2, localizing it to the nucleus and preventing SIAH1-mediated ubiquitination of HIPK2. Related references: PMID:33591310
  • Subcellular localization includes nucleus, PML bodies, cytoplasm, and stress granules, reflecting its dynamic role in signaling.
  • Highly expressed in heart, muscle, and kidney; down-regulated in certain thyroid and breast tumors.
  • Alternatively spliced isoforms produce proteins of ~101–131 kDa, consistent with observed multiple bands in western blot.
  • Key post-translational modifications include phosphorylation and ubiquitination; implicated in host-virus interactions.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide mapping to residues 100-200 of human HIPK2; verify reactivity in target species and tissue.
  • For Western blot, multiple bands around 101–131 kDa may be observed due to alternative splicing; include appropriate controls.
  • In immunofluorescence, anticipate nuclear and possibly cytoplasmic localization; optimize fixation and permeabilization conditions.
  • For ELISA, consider pairing with a detection antibody targeting a distinct epitope; validate in the relevant sample matrix.
