Biological Background: HIP1R Function and Localization
- HIP1R, also known as huntingtin-interacting protein 12 (HIP12), is a 119 kDa protein encoded by the HIP1R gene (GeneID: 9026) and belongs to the HIP1 family.
- It functions as a component of clathrin-coated pits and vesicles, linking the endocytic machinery to the actin cytoskeleton via its ENTH domain, which binds 3-phosphoinositides.
- HIP1R may promote cell survival by stabilizing receptor tyrosine kinases following ligand-induced endocytosis.
- The protein localizes to the cytoplasm, particularly in the perinuclear region, and associates with the endomembrane system and clathrin-coated vesicle membranes.
- Expression is detected in brain, heart, kidney, pancreas, and liver, but not in lung or placenta, suggesting tissue-specific roles in endocytosis and signaling.
- Post-translational modifications include acetylation and phosphorylation, and the protein features an ENTH domain, coiled-coil regions, and actin-binding motifs.
Experimental Guidance and Technical Tips
- The immunogen was designed against amino acids 300–600 of human HIP1R (NP_003950.1), spanning a region rich in potential epitopes; this may enhance specificity for both endogenous and recombinant forms.
- For Western blot, expect a band at approximately 119 kDa; consider using positive control lysates from brain or heart tissues, or cell lines known to express HIP1R.
- In immunofluorescence/immunocytochemistry, HIP1R displays a punctate cytoplasmic pattern due to its association with clathrin-coated vesicles; optimize fixation and permeabilization accordingly.
- An ELISA protocol can be developed using this antibody; validate the assay with known concentrations of recombinant HIP1R protein.
