Biological Background: HINT1 Function and Localization
- Histidine triad nucleotide-binding protein 1 (HINT1) is a member of the HIT superfamily and is also known as protein kinase C inhibitor 1 (PKCI-1) and adenosine 5'-monophosphoramidase.
- Functions as a hydrolase that catalyzes the hydrolysis of purine nucleotide phosphoramidates, such as adenosine 5'-monophosphoramidate (AMP-NH2), to AMP and ammonia. It also hydrolyzes various aminoacyl-AMP and morpholidate substrates. Related references: PMID:15703176, PMID:16835243, PMID:17217311, PMID:17337452, PMID:22329685, PMID:23614568, PMID:30772266
- Acts as a desumoylating isopeptidase, deconjugating SUMO1 from proteins such as RGS17 and RANGAP1. Related references: PMID:31088288
- Serves as a scaffolding protein that modulates transcriptional activation by the LEF1/TCF1-CTNNB1 and MITF-CTNNB1 complexes. Related references: PMID:16014379, PMID:22647378
- Modulates p53/TP53 protein levels and p53-mediated apoptosis. Related references: PMID:16835243
- Modulates proteasomal degradation of target proteins by the SCF (SKP2-CUL1-F-box protein) E3 ubiquitin-protein ligase complex. Related references: PMID:19112177
- Localizes to both the cytoplasm and nucleus, consistent with its diverse enzymatic and regulatory roles.
- Ubiquitously expressed across tissues, reflecting its fundamental cellular functions.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length HINT1 protein (amino acids 1–126), suggesting the antibody may recognize linear and conformational epitopes.
- For Western blot, the predicted molecular weight is ~14 kDa. Use appropriate gel percentage and membrane transfer conditions to retain this small protein.
- In IF/ICC, optimize fixation (e.g., 4% PFA versus methanol) and permeabilization, as detection of nuclear and cytoplasmic pools may vary.
- Validate the antibody in the relevant sample system (human, mouse, or rat) using known positive and negative control lysates or cells, as cross-reactivity may differ by application.
- For ELISA, consider direct coating of recombinant HINT1 protein or a sandwich format; confirm the antibody’s reactivity with the coating antigen under your buffer conditions.
