Biological Background: HIF1β/ARNT Function and Localization
- HIF1β/ARNT (Aryl hydrocarbon receptor nuclear translocator) is a transcription factor encoded by the ARNT gene (also known as BHLHE2). It functions as an essential heterodimerization partner for multiple basic helix-loop-helix PER-ARNT-SIM (bHLH-PAS) proteins, mediating responses to environmental stimuli and developmental cues.
- Forms heterodimers with the aryl hydrocarbon receptor (AHR) to regulate xenobiotic metabolism by binding to xenobiotic response elements (XREs) in target gene promoters. Related references: PMID:28396409, PMID:34521881
- Partners with hypoxia-inducible factor alpha subunits (HIF1A, EPAS1, NPAS1, NPAS3) to form hypoxia-inducible transcription factors that bind hypoxia response elements (HREs), activating genes involved in angiogenesis, erythropoiesis, and metabolic adaptation to low oxygen. Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 420-485 near the C-terminus of human HIF1β/ARNT (NP_001659.1). This region is expected to be accessible in native conformations, but epitope masking may occur in some complexes; validate in the relevant sample system.
- Primarily recommended for Western blot (WB) and ELISA applications. For WB, the predicted molecular weight is approximately 87 kDa; however, post-translational modifications may cause band shifts. Use appropriate positive and negative controls.
- Since the antibody cross-reacts with human and mouse, it is suitable for studies in both species. Verify reactivity in other species if needed.
- For ELISA, consider pairing with an appropriate capture antibody or using the recommended assay protocol. Titrate for optimal signal-to-noise.
