- The immunogen is a synthetic peptide corresponding to amino acids 400–500 of human HIF1α (NP_001521.1), which may contribute to the antibody's cross-reactivity with mouse and rat orthologs.
- For Western blot (WB), HIF1α typically migrates at 93–96 kDa; additional bands corresponding to splice isoforms or modified forms may be observed. Validate in the relevant sample system, preferably using hypoxic stimuli to increase target expression.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimization of fixation, antigen retrieval, and signal amplification is recommended. Consider including hypoxic tissue samples as positive controls.
