Biological Background: HIF1α Function and Localization

  • It forms a heterodimer with ARNT and binds to hypoxia response elements (HREs) containing the core sequence 5'-TACGTG-3' in target gene promoters.   • The antibody was raised against a synthetic peptide and is expected to recognize total HIF1α protein across human and mouse species. Confirm reactivity in your specific experimental model.
  • For Western blot, anticipate bands at approximately 83, 93, or 96 kDa depending on isoform expression and post-translational modifications; use appropriate molecular weight markers and hypoxia-treated positive controls to verify detection.
  • In immunofluorescence/ICC, optimize fixation and permeabilization to preserve both cytoplasmic and nuclear localization signals; induction of HIF1α stabilization by hypoxia mimetics or low oxygen incubation is recommended prior to staining.
  • For ELISA applications, validate the antibody pair with recombinant HIF1α or hypoxia-treated cell lysates to ensure linear detection range and sensitivity.
