- HHLA2 (also known as B7-H7, B7y, B7-H5) is a member of the B7 family of immune regulatory ligands, characterized by an immunoglobulin domain, disulfide bonds, and glycosylation motifs.
- It functions as a costimulatory molecule on antigen-presenting cells, interacting with TMIGD2 on T-cells to enhance TCR-mediated activation, proliferation, and cytokine production via an AKT-dependent signaling cascade.
- HHLA2 is a single-pass type I transmembrane glycoprotein localized on the cell surface, with a signal peptide, extracellular Ig-like domains, a transmembrane helix, and a cytoplasmic tail.
- It is expressed in a variety of tissues, including colon, kidney, testis, lung, and pancreas, and at lower levels in small intestine, liver, and skeletal muscle. In immune cells, it is highly expressed in B-cells, dendritic cells, and macrophages, but not detected in T-cells.
- The protein undergoes alternative splicing, producing multiple isoforms that may differ in functional properties.
- Key post-translational modifications include N-linked glycosylation and disulfide bond formation, which are critical for its structural stability and ligand-receptor interactions.
