Biological Background: HGF Function and Localization
- Hepatocyte growth factor (HGF), also known as scatter factor (SF) or hepatopoietin-A, is encoded by the HGF gene and plays a central role in liver regeneration and tissue repair.
- HGF is a potent mitogen for mature parenchymal hepatocytes and acts as a growth factor for a broad spectrum of tissues and cell types. Related references: PMID:20624990
- It serves as the activating ligand for the receptor tyrosine kinase MET, promoting receptor dimerization and downstream MAPK signaling. Related references: PMID:15167892, PMID:20977675
- The protein is synthesized as an inactive single-chain precursor and undergoes proteolytic cleavage to generate a biologically active heterodimer composed of an alpha and a beta chain linked by a disulfide bond.
- HGF contains multiple structural domains, including kringle domains and a serine protease homology region; however, the serine protease domain lacks catalytic activity.
- Mutations in HGF are associated with non-syndromic hearing loss (DFNB39), highlighting its importance beyond hepatic function.
- Alternative splicing yields multiple isoforms, and the protein undergoes several post-translational modifications such as glycosylation and formation of pyrrolidone carboxylic acid.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 106–160 of human HGF, a region that is highly conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blotting, a band around 83 kDa (full-length precursor) is expected; additional bands corresponding to processed forms may also be detected. Consider validating the detection of cleaved forms in your specific sample system.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization conditions should be optimized for the target tissue. The antibody may be suitable for visualizing HGF in liver, placenta, and other expressing tissues.
- Given the polyclonal nature, lot-to-lot consistency should be verified for critical quantitative applications. We recommend titrating the antibody from 1:500 to 1:2000 for initial Western blot experiments.
