Biological Background: HEYL Function and Localization
- HEYL (Hairy/enhancer-of-split related with YRPW motif-like protein), also designated HEY3, HRT3, HESR3, and bHLHb33, is a basic helix-loop-helix (bHLH) transcription factor.
- Acts as a transcriptional repressor by binding to the canonical E box sequence 5'-CACGTG-3', thereby regulating gene expression downstream of Notch signaling.
- Functions as an effector of Notch signaling, potentially required for proper cardiovascular development.
- Represses transcription mediated by the cardiac transcriptional activators GATA4 and GATA6.
- Primarily localizes to the nucleus, consistent with its role as a DNA-binding transcription factor.
- Belongs to the HEY family of transcriptional repressors, which are characterized by a bHLH domain and a conserved YRPW motif.
Experimental Guidance and Technical Tips
- The immunogen, a recombinant fusion protein corresponding to the N-terminal region (aa 1-100) of human HEYL, may generate antibodies recognizing the native protein and possibly other family members; verify specificity.
- For WB, the predicted molecular weight of HEYL is approximately 35 kDa; however, post-translational modifications or isoform variants might affect migration. Include appropriate controls.
- In ELISA, the polyclonal nature of this antibody could be advantageous for capturing the target; consider optimizing coating and detection conditions.
- Cross-reactivity is reported for human and mouse; for other species, validate reactivity experimentally.
