Biological Background: Hexokinase II Function and Localization
- Hexokinase-2 (HK2), also known as HKII, HXK2, hexokinase type II, hexokinase-B, and muscle form hexokinase, is a member of the hexokinase family and is a 102 kDa protein encoded by the HK2 gene (Gene ID: 3099).
- Catalyzes the phosphorylation of hexoses, such as D-glucose and D-fructose, to hexose 6-phosphates, mediating the first committed step of glycolysis (PubMed:23185017, PubMed:26985301, PubMed:29298880). Related references: PMID:23185017 PMID:26985301 PMID:29298880
- Plays a key role in maintaining the integrity of the outer mitochondrial membrane by preventing the release of apoptogenic molecules from the intermembrane space, thereby inhibiting apoptosis (PubMed:18350175). Related references: PMID:18350175
- Predominantly expressed in insulin-responsive tissues such as skeletal muscle.
- Localizes to the mitochondrial outer membrane and the cytosol.
- Post-translational modifications include acetylation; it contains ATP-binding domains and acts as an allosteric enzyme (UniProt keywords: Acetylation, Allosteric enzyme, ATP-binding).
Experimental Guidance and Technical Tips
- The immunogen (recombinant fragment spanning amino acids 1-120) targets the N-terminal region of human Hexokinase II; consider its potential to detect various isoforms or cleavage products.
- For Western blot (WB), the predicted molecular weight is ~102 kDa; optimize gel percentage and transfer conditions for high-molecular-weight proteins.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), include appropriate antigen retrieval and permeabilization steps; validate in tissues with known HK2 expression (e.g., skeletal muscle, heart).
- Owing to cross-reactivity with mouse and rat, the antibody can be applied across human, mouse, and rat samples in WB, IHC, IF, and ELISA; validate in the relevant species system.
