Biological Background: Hexokinase II Function and Localization

  • Hexokinase-2 (HK2, gene HK2) is the predominant hexokinase isozyme expressed in insulin-responsive tissues such as skeletal muscle and plays a central role in cellular hexose metabolism.
  • HK2 catalyzes the ATP-dependent phosphorylation of D-glucose to D-glucose 6-phosphate, the first committed step of glycolysis, as well as the phosphorylation of other hexoses like D-fructose. Related references: PMID:29298880, PMID:23185017, PMID:26985301
  • The enzyme binds to the mitochondrial outer membrane, preserving membrane integrity and preventing the release of pro-apoptotic molecules, thereby inhibiting apoptosis. Related references: PMID:18350175
  • HK2 shuttles between the cytosol and the mitochondrial outer membrane, a distribution influenced by metabolic signals and post-translational modifications such as acetylation.
  • As an allosteric enzyme, HK2 is feedback-inhibited by glucose-6-phosphate; its high expression in many cancers supports the Warburg effect by maintaining elevated glycolytic flux.
  • The full-length human HK2 protein has a molecular weight of approximately 102 kDa and contains ATP- and nucleotide-binding domains essential for its kinase activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal amino acids 1–120 of human HK2, a region well conserved across human, mouse, and rat, consistent with the observed cross-reactivity.
  • For Western blotting, expect a band at ~102 kDa; optimize gel percentage and transfer conditions for high–molecular-weight proteins.
  • In ELISA applications, use recombinant HK2 protein or lysates from HK2-rich tissues (e.g., skeletal muscle, cancer cell lines) as positive controls.
  • Validate antibody performance in your specific experimental system using appropriate positive and negative controls, and consider testing multiple dilutions.
