Biological Background: HES1 Function and Localization
- Transcription factor HES-1, also known as Class B basic helix-loop-helix protein 39 (bHLHb39), Hairy and enhancer of split 1, or Hairy-like protein (hHL), is a member of the bHLH transcription factor family.
- HES-1 acts as a transcriptional repressor of genes that require a bHLH protein for their transcription. It binds DNA on N-box motifs (5'-CACNAG-3') with high affinity and on E-box motifs (5'-CANNTG-3') with low affinity.
- It functions as a negative regulator of myogenesis by inhibiting the activities of MYOD1 and ASH1.
- HES-1 plays a role in the cellular response to DNA cross-link damage, being required for the stability and nuclear localization of FA core complex proteins, as well as for FANCD2 monoubiquitination.
- The protein is localized to the nucleus, consistent with its role as a transcription factor.
- According to UniProt keywords, HES-1 is subject to ubiquitin conjugation (Ubl conjugation), which may regulate its activity or stability in DNA damage signaling.
Experimental Guidance and Technical Tips
- For Western blot detection, nuclear-enriched lysates are recommended due to the predominant nuclear localization of HES-1; the predicted molecular weight is ~30 kDa.
- In immunohistochemistry (IHC-P), antigen retrieval conditions should be optimized for the specific tissue and fixation method; positive nuclear staining is expected.
- For immunoprecipitation, consider testing different elution buffers to maintain antigen-antibody complex integrity.
- The immunogen corresponds to the C-terminal region (aa 181-280); thus, the antibody may recognize both full-length and potential truncated forms—validate in the relevant sample system.
- Cross-reactivity with mouse and rat is reported; however, we recommend performing a sequence alignment if using in other species.
