Biological Background: HER2/ErbB2 Function and Localization

  • HER2 (Receptor tyrosine-protein kinase erbB-2) is a member of the epidermal growth factor receptor (EGFR/ErbB) family, encoded by the ERBB2 gene. It is also known as p185erbB2, CD340, HER-2/neu, and proto-oncogene Neu.
  • Functions as a protein tyrosine kinase that forms heterodimers with other ErbB family members, acting as the preferred dimerization partner. It plays a critical role in signal transduction pathways regulating cell growth, differentiation, and survival.
  • In the cytoplasm, HER2 regulates microtubule stabilization and outgrowth via the MEMO1-RHOA-DIAPH1 signaling pathway, which leads to inhibition of GSK3B and membrane association of APC and CLASP2, crucial for microtubule capture.
  • In the nucleus, HER2 functions as a transcriptional regulator, directly binding to the PTGS2/COX-2 promoter to activate transcription. It also regulates CDKN1A expression through STAT3 and SRC and stimulates rRNA synthesis by RNA Pol I, promoting protein synthesis and cell growth.
  • Subcellular localization is dynamic: primarily at the cell membrane, but also found in early endosomes, cytoplasm (perinuclear region), nucleus, and cell projections like ruffles.
  • Expressed in various tumor tissues, including primary breast tumors and cancers of the small bowel, esophagus, kidney, and mouth, marking it as a critical oncogene and therapeutic target.
  • Post-translational modifications include phosphorylation, glycosylation, and disulfide bond formation, which are essential for its activation and function as a transmembrane receptor kinase.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 23-652) of human HER2, making this antibody suitable for detecting surface-expressed HER2 in live, non-permeabilized cells by flow cytometry.
  • For intracellular staining, consider gentle permeabilization protocols to access nuclear or cytoplasmic pools of HER2 while preserving epitope integrity.
  • Titrate the antibody to determine the optimal concentration for FC assays, as excessive antibody may increase background or cause aggregation.
  • Always include appropriate isotype controls and single-stain controls to ensure accurate compensation and gating.
  • Protect the conjugate from light and avoid freeze-thaw cycles to maintain fluorophore stability and antibody performance.
