Biological Background: Heparanase Function and Localization
- Heparanase (HPSE, also known as HPA1, HPR1, HSE1, HPSE1) is an endo-β-glucuronidase that specifically cleaves heparan sulfate proteoglycans (HSPGs) into smaller fragments, playing a key role in extracellular matrix (ECM) degradation and remodeling.
- The enzyme exhibits pH-dependent activity, being essentially inactive at neutral pH but highly active under acidic conditions, such as those encountered during tumor invasion, inflammation, and wound healing.
- Heparanase is initially synthesized as a 65 kDa precursor that undergoes proteolytic processing to yield a mature, active heterodimer composed of 8 kDa and 50 kDa subunits; the mature enzyme localizes primarily to lysosomes and can be secreted.
- Subcellular fractionation and immunohistochemistry studies show Heparanase at the lysosome membrane, in the nucleus, and as a secreted protein, suggesting versatile roles in both intracellular and extracellular microenvironments.
- Tissue expression is prominent in the placenta and spleen, with lower levels in lymph node, thymus, peripheral blood leukocytes, bone marrow, endothelial cells, fetal liver, and various tumor tissues; within hair follicles, it is specifically detected in the inner root sheath layers.
- Beyond its enzymatic function, Heparanase modulates cell adhesion, promotes AKT1/PKB phosphorylation via lipid rafts, upregulates VEGF-mediated angiogenesis through SRC activation, and acts as a procoagulant by enhancing factor X activation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 36–275) corresponds to a region that likely excludes the N-terminal signal peptide, potentially targeting the mature form of Heparanase; consider verifying reactivity with both the pro-form (~65 kDa) and processed subunits (50 kDa and 8 kDa) in your assay conditions.
- For western blotting, prepare lysates at neutral pH and consider running a positive control from placenta or spleen tissue, where HPSE expression is high; a band near 61–65 kDa is expected.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), test the antibody on paraffin-embedded or frozen sections of placenta, spleen, or tumor xenografts, which strongly express Heparanase, and include an isotype control.
- For ELISA applications, use as a capture or detection antibody after confirming pairing compatibility; recombinant Heparanase protein (e.g., the immunogen) can serve as a standard.
