- Hemoglobin subunit alpha (alpha-globin) is the alpha chain of hemoglobin, a heterotetramer of two alpha and two beta chains, encoded by the HBA1 and HBA2 genes.
- Its primary function is oxygen transport from the lungs to peripheral tissues.

- It is predominantly expressed in red blood cells.
- The protein undergoes various post-translational modifications including acetylation, glycation, and phosphorylation.
- It contains a heme group with an iron atom essential for oxygen binding, and mutations are associated with hereditary hemolytic anemia.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human HBA1; the antibody may recognize both HBA1 and the nearly identical HBA2 (differ by one amino acid). Consider testing cross-reactivity if isoform distinction is critical.
- For Western blot, the expected band is around 15 kDa. Use reducing conditions as hemoglobin is a heterotetramer.
- For ELISA, validate with recombinant HBA1 or red blood cell lysates.
- Since HBA1 is highly abundant in red blood cells, consider appropriate sample preparation to avoid overloading.
- The antibody is validated in human samples; cross-reactivity with other species is not tested based on provided data. If using other species, validate by sequence alignment.
