Biological Background: Heme Oxygenase 1 Function and Localization
- Heme oxygenase 1 (HO-1) is encoded by the HMOX1 gene and is also known as HSP32; it is a member of the heme oxygenase family and functions as an endoplasmic reticulum-anchored enzyme.
- HO-1 catalyzes the oxidative cleavage of heme at the alpha-methene bridge, producing carbon monoxide (CO), biliverdin IXalpha, and ferrous iron, a critical step in heme catabolism and iron recycling (PubMed:11121422, PubMed:19556236, PubMed:7703255). Related references: PMID:11121422 PMID:19556236 PMID:7703255
- The cytoprotective effects of HO-1 are mediated by its ability to degrade pro-oxidant free heme, thereby preventing heme-induced apoptosis (PubMed:20055707). Related references: PMID:20055707
- HO-1 localizes to the endoplasmic reticulum membrane through a C-terminal transmembrane helix, positioning it for optimal interaction with heme substrates.
- Post-translational modifications such as phosphorylation, along with its heme and iron binding properties, classify HO-1 as a phosphoprotein, metalloprotein, and oxidoreductase.
- During SARS-CoV-2 infection, HO-1 promotes ORF3A-mediated autophagy, though it is not required for ORF3A-induced reticulophagy.
- In renal cancer, HO-1 protein expression is upregulated compared to normal tissue, hinting at a role in tumorigenesis.
Experimental Guidance and Technical Tips
- Given the immunogen corresponds to mouse Hmox1 (aa 1-266), the antibody is expected to recognize both human and mouse HO-1; researchers should validate specificity in the intended species via endogenous positive controls.
- For Western blot, a band near 33 kDa is predicted; consider using whole-cell lysates from heme-treated or stress-induced cells to enhance detection.
- When performing immunoprecipitation (IP), ensure antibody compatibility with the buffer conditions and confirm capture by immunoblotting with a distinct anti-HO-1 antibody.
- For immunofluorescence (IF) and immunohistochemistry (IHC), include appropriate fixation and permeabilization steps; use tissue sections or cells known to express HO-1 (e.g., renal cancer tissue or heme-stimulated macrophages) as positive controls.
- In ELISA applications, optimize coating antigen and detection antibody concentrations using recombinant HO-1 protein standards.
