Biological Background: HEC1/NDC80 Function and Localization
- HEC1 (Highly Expressed in Cancer protein 1), also known as NDC80, HEC, TID3, or KNTC2, is a kinetochore protein encoded by the NDC80 gene in humans. It is a component of the essential NDC80 complex required for chromosome segregation and spindle checkpoint activity. Related references: PMID:12351790 PMID:14654001 PMID:14699129
- The NDC80 complex is critical for kinetochore integrity and the organization of stable microtubule binding sites in the outer kinetochore plate. Related references: PMID:15548592 PMID:30409912
- HEC1 synergistically enhances the affinity of the SKA1 complex for microtubules and may allow the NDC80 complex to track depolymerizing microtubules. Related references: PMID:23085020
- The protein plays an essential role in chromosome congression and the end-on attachment of kinetochores to spindle microtubules, ensuring proper chromosome alignment during mitosis. Related references: PMID:23891108 PMID:25743205
- HEC1 localizes to the nucleus, specifically at the centromere and kinetochore regions of chromosomes. It is associated with cell cycle, cell division, centromere, chromosome, kinetochore, and mitosis keywords.
- Post-translational modifications include acetylation and phosphorylation, which regulate its function during mitotic progression.
- The full-length protein has a calculated molecular weight of approximately 74 kDa, consistent with the observed band in western blot.
Experimental Guidance and Technical Tips
- The immunogen comprises a recombinant fragment (aa 343-642) of human HEC1, covering the C-terminal region known for microtubule-binding activity. This design promotes recognition of native protein isoforms.
- For WB, consider using whole cell lysates from human or mouse cell lines (e.g., HeLa, NIH/3T3) to validate detection of the 74 kDa band.
- In ELISA, the antibody may be used as a capture or detection reagent; titrate appropriately for optimal signal-to-noise ratio in your specific assay format.
- Since HEC1 is a key mitotic regulator, synchronize cells at metaphase if stronger signal is needed for co-localization or functional studies.
