Biological Background: HE4/WFDC2 Function and Localization
- HE4/WFDC2 is a secreted WAP four-disulfide core domain protein (UniProt Q14508), also known as Epididymal secretory protein E4 and Major epididymis-specific protein E4.
- It plays a critical role in lung development, being required for terminal differentiation of bronchiole club cells and formation of airway barrier mechanisms, including lamellar body formation in type II alveolar epithelial cells and cilia formation.
- The protein is essential for type I alveolar epithelial cell and vascular endothelial development in the lungs, mediating blood-air barrier exchange postnatally.
- HE4/WFDC2 belongs to the WAP domain family, characterized by a conserved four-disulfide core, and is classified as a putative protease inhibitor with activity against aspartic, serine, and thiol proteases.
- It is expressed in a wide range of tissues including lung, trachea, kidney, breast, fallopian tubes, endometrium, cervix, epididymis, and prostate.
- Alternative splicing generates multiple isoforms, and the protein is glycosylated, with a predicted molecular weight of approximately 13 kDa.
- The gene is associated with disease variants, highlighting its clinical relevance in conditions such as ovarian cancer (often used as a biomarker).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning residues 45-124, which may be useful for detecting both full-length and certain isoforms of HE4/WFDC2.
- For Western blot, consider using reducing conditions to account for the multiple disulfide bonds characteristic of WAP domain proteins.
- In immunofluorescence/ICC, permeabilization may be required due to the secreted nature of the target; optimize fixation and permeabilization protocols for your cell type.
- Validate antibody reactivity in your specific cell or tissue lysates, as expression levels can vary; positive controls may include lung, epididymis, or ovarian cancer cell lines.
