Biological Background: HDAC6 Function and Localization
- HDAC6 (histone deacetylase 6) is a cytoplasmic enzyme that deacetylates a wide range of non-histone substrates, including α-tubulin, cortactin, and peroxiredoxins, thereby regulating microtubule dynamics, cell motility, and redox homeostasis. Related references: PMID:12606581, PMID:19893491
- It plays a central role in microtubule-dependent cell motility by deacetylating α-tubulin, which destabilizes dynamic microtubules and is required for cilia disassembly. Related references: PMID:12486003, PMID:27737934
- HDAC6 acts as an adapter that recognizes polyubiquitinated misfolded proteins and targets them to the aggresome, facilitating their clearance by autophagy. Related references: PMID:22819792
- The enzyme deacetylates the antiviral protein RIG-I, a modification required for viral RNA detection, and also regulates PRDM16 to control brown adipocyte function. Related references: PMID:26746851, PMID:35075301
- HDAC6 localizes predominantly to the cytoplasm but is also found in the nucleus, centrosome, basal body, and neuronal projections including dendrites, axons, and cilia.
- In mice, HDAC6 is expressed in cortical neurons, Purkinje cells, and keratinocytes (at protein level).
- The protein contains two catalytic deacetylase domains and a C-terminal ubiquitin-binding zinc-finger domain, classifying it as a class IIb HDAC with roles in transcription regulation and chromatin remodeling.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 836–1037) of mouse HDAC6, which includes part of the second deacetylase domain and the ubiquitin-binding zinc finger. This design may favor detection of full-length HDAC6 and potentially specific isoforms.
- This rabbit monoclonal antibody offers batch-to-batch consistency and has been qualified for WB, IF-P, IHC-P, and ELISA by the supplier.
- Given the large predicted protein size (~126 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6–8%) and extended transfer times for western blotting. A high-molecular-weight marker is recommended.
- For immunofluorescence or immunohistochemistry, optimize permeabilization and fixation conditions, as HDAC6 is both cytoplasmic and nuclear. Microtubule-associated staining may benefit from anti-tubulin co-staining.
- Cross-reactivity has been confirmed in mouse and rat; additional species should be validated independently.
