- Through histone deacetylation, HDAC5 contributes to transcriptional regulation, cell cycle progression, and developmental processes, typically as part of large multiprotein complexes.
- HDAC5 represses transcription of the myocyte enhancer factor MEF2C; during muscle differentiation, it shuttles to the cytoplasm, thereby derepressing myogenic genes.
- In breast cancer, HDAC5 is involved in MTA1-mediated epigenetic silencing of ESR1 (estrogen receptor alpha) expression.
