Biological Background: HDAC3 Function and Localization

  • HDAC3 is a histone deacetylase that removes acetyl groups from lysine residues on core histones (H2A, H2B, H3, H4) and non-histone substrates, playing a central role in epigenetic repression, transcriptional regulation, cell cycle progression, and development. Related references: PMID:21030595, PMID:21444723, PMID:23911289

  • It operates within large multiprotein complexes such as the N-CoR repressor complex, which activates its deacetylase activity, and participates in BCL6-mediated transcriptional repression by deacetylating H3K27 on enhancer elements. Related references: PMID:23911289, PMID:22230954

  • Regulates both the activation and repression phases of the circadian clock in a deacetylase activity-independent manner, involving BMAL1 ubiquitination and CRY1/BMAL1 interaction (By similarity).

  • Contributes to endothelial cell survival under oxidative stress by activating HMOX1 transcription via XBP1 isoform 1 and a PI(3)K/mTORC2/Akt-dependent signaling pathway. Related references: PMID:25190803

  • Deacetylates a range of non-histone proteins, including KAT5, MEF2D, MAPK14, RARA, and STAT3, thereby extending its regulatory functions beyond chromatin. Related references: PMID:15653507, PMID:21030595, PMID:21444723

  • Acts as a corepressor of RARA, mediating its deacetylation and inhibiting RARE DNA element binding; involved in the inflammatory response by repressing microRNA-10a expression. Related references: PMID:28167758

  • Additionally functions as a protein-lysine deacylase, capable of removing crotonyl, lactoyl, 2-hydroxyisobutyryl, and isonicotinyl groups; catalyzes decrotonylation of MAPRE1/EB1 and delactylation of NBN/NBS1, the latter inhibiting DNA double-strand breaks via homologous recombination. Related references: PMID:28497810, PMID:29192674, PMID:34608293, PMID:38961290

  • Localizes primarily in the nucleus but also found in the chromosome, cytoplasm, and cytosol; it is widely expressed across tissues. Post-translational modifications include phosphorylation and ubiquitin conjugation, and alternative splicing generates multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 329–428 of human HDAC3; consider confirming antibody specificity in your experimental model using relevant controls.
  • Validated applications include Western blot (WB), immunoprecipitation (IP), and ELISA. The predicted molecular weight is 49 kDa; optimize gel electrophoresis and transfer conditions for WB.
  • Cross-reactivity is observed with human, mouse, and rat samples, suggesting conserved epitope recognition; verify performance in other species if required.
  • For IP, titrate the antibody concentration and include appropriate negative and positive controls to ensure pull-down specificity.
  • Given HDAC3’s association with various corepressor complexes, this antibody may be suitable for co-immunoprecipitation studies to analyze protein-protein interactions.
