Biological Background: HDAC3 Function and Localization
- Acts as a histone deacetylase, catalyzing the deacetylation of lysine residues on core histones (H2A, H2B, H3, H4) and various non-histone proteins, leading to epigenetic repression and playing critical roles in transcriptional regulation, cell cycle progression, and developmental events. Related references: PMID:21030595, PMID:21444723, PMID:23911289
- Functions within large multiprotein complexes such as the N-Cor repressor complex and participates in BCL6-mediated transcriptional repression by deacetylating H3K27 on enhancer elements, antagonizing EP300 acetyltransferase activity. Related references: PMID:23911289, PMID:22230954
- Deacetylates non-histone substrates, including KAT5, MEF2D, MAPK14, RARA, and STAT3, thereby modulating their activities in diverse cellular signaling pathways. Related references: PMID:15653507, PMID:21030595, PMID:21444723
- Also acts as a protein-lysine deacylase, catalyzing the removal of crotonyl, lactoyl, 2-hydroxyisobutyryl, and isonicotinyl modifications; for instance, decrotonylates MAPRE1 and delactylates NBN, affecting DNA double-strand break repair. Related references: PMID:28497810, PMID:29192674, PMID:34608293, PMID:38961290
- Contributes to circadian clock regulation in a deacetylase-independent manner, promoting BMAL1 ubiquitination during activation and blocking CRY1/2 degradation during repression; also influences hepatic lipid metabolism gene expression.
- Localizes primarily in the nucleus and on chromosomes, with additional presence in the cytoplasm and cytosol, consistent with its transcriptional regulatory and shuttling functions.
- Widely expressed across multiple tissues, reflecting its fundamental role in epigenetic control.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide covering amino acids 329–428 of human HDAC3 (C-terminal region). When extending the use to other species, confirm sequence homology in this region.
- Optimize antibody concentration for each assay. For Western blotting, consider a starting dilution of 1:500–1:2000, and include appropriate positive and negative controls. For immunohistochemistry, antigen retrieval may improve detection.
- This antibody has shown reactivity with human, mouse, and rat HDAC3. If used with other species, perform cross-reactivity validation.
- The predicted molecular weight is 49 kDa; verify the band size in your experimental system, as post-translational modifications may cause subtle shifts.
