Biological Background: HDAC2 Function and Localization
- HDAC2 (histone deacetylase 2), also referred to as protein deacylase HDAC2, is a member of the histone deacetylase family.
- Catalyzes the deacetylation of lysine residues on the N-terminal tails of core histones (H2A, H2B, H3, and H4), leading to epigenetic repression and playing a critical role in transcriptional regulation, cell cycle progression, and development. Related references: PMID:28497810
- Forms transcriptional repressor complexes via association with MAD, SIN3, YY1, and N-COR, contributing to gene silencing. Related references: PMID:12724404
- Serves as a component of the histone deacetylase NuRD complex, which participates in chromatin remodeling, and the SIN3B complex that counteracts histone acetyltransferase EP300 through recognition of H3K27ac marks and HDAC2 activity. Related references: PMID:16428440, PMID:28977666, PMID:37137925
- Deacetylates non-histone proteins, including TSHZ3 (regulating its transcriptional repressor activity) and mediates MTA1-dependent repression of TFF1 and CDKN1A. Related references: PMID:19343227, PMID:21965678
- Functions as a protein-lysine deacylase, removing crotonyl, lactyl, and 2-hydroxyisobutyryl modifications from lysine residues. Related references: PMID:28497810, PMID:29192674, PMID:35044827
- Localizes to both the nucleus and cytoplasm, consistent with its roles in chromatin regulation and potential cytoplasmic functions.
- Widely expressed across tissues, with relatively lower levels detected in brain and lung.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning amino acids 389–488 of human HDAC2; based on sequence homology, this antibody is expected to cross-react with mouse and rat HDAC2 as indicated, but confirm in your specific samples.
- In Western blot, a primary band around 55 kDa is anticipated; however, electrophoretic mobility may vary slightly depending on cell type and post-translational modifications.
- For chromatin immunoprecipitation (ChIP), optimize crosslinking and antibody concentration, and verify enrichment by qPCR targeting known HDAC2-regulated promoters (e.g., TFF1, CDKN1A).
- For immunoprecipitation, include appropriate negative controls (e.g., IgG isotype control) to ensure specificity.
