- Post-translational modifications include N-linked glycosylation, disulfide bonds stabilizing the prefusion structure, and palmitoylation anchoring the protein in the lipid bilayer.

- The immunogen peptide lies within the S2 subunit (aa 782–881); therefore, the antibody is expected to recognize both full-length Spike (~150 kDa) and the S2 cleavage product under reducing conditions.
- For WB, consider using HCoV-OC43-infected cell lysate as a positive control; glycosylation may cause the band to migrate at a higher apparent molecular weight.
- Due to the polyclonal nature, the antibody may provide broad epitope coverage in ELISA; however, cross-reactivity with other coronaviruses should be assessed empirically.
- As a transmembrane glycoprotein, Spike requires appropriate detergents for extraction; avoid prolonged boiling of samples to prevent aggregation.
