Biological Background: Spike glycoprotein Function and Localization
- Spike glycoprotein (S protein, E2, peplomer protein) is the major envelope protein of HCoV-NL63, mediating viral entry into host cells.
- The S1 subunit binds to the host receptor angiotensin-converting enzyme 2 (ACE2) to initiate infection. Related references: PMID:15897467 PMID:19901337 PMID:29142129
- The S2 subunit is a class I viral fusion protein that undergoes dramatic conformational changes to fuse viral and host membranes.
- The protein transitions through at least three states: pre-fusion native, pre-hairpin intermediate, and post-fusion hairpin, with coiled-coil heptad repeats forming a trimer-of-hairpins structure.
- Located on the virion membrane and in host endoplasmic reticulum-Golgi intermediate compartment membranes.
- Contains disulfide bonds and is heavily glycosylated, typical of viral envelope proteins, and functions in viral attachment and entry.

Experimental Guidance and Technical Tips
- As a polyclonal antibody generated against a peptide immunogen in the S2 region, it may recognize both native and denatured Spike protein; verify by pilot experiments.
- For Western blotting under reducing conditions, expect a band at approximately 150 kDa; run appropriate molecular weight markers.
- For ELISA applications, consider using as a coating antibody or detection antibody paired with a suitable secondary antibody; validate with HCoV-NL63 antigen.
- Since cross-reactivity is reported only with HCoV-NL63, do not assume detection of other coronaviruses without experimental confirmation.
