Biological Background: HAX1 Function and Localization
- HAX1 (HCLS1-associated protein X-1), also known as HS1-associating protein X-1 (HAX-1) and HS1-binding protein 1 (HSP1BP-1), is an actin cytoskeleton regulatory protein that recruits the Arp2/3 complex to the cell cortex and modulates KCNC3 potassium channel inactivation (PubMed:26997484). Related references: PMID:26997484
- It promotes cell migration through GNA13 signaling and participates in clathrin-mediated endocytosis, including the internalization of ABC transporters such as ABCB11.
- HAX1 plays an anti-apoptotic role by inhibiting caspase-9 and caspase-3, thereby supporting cell survival.
- It contributes to the regulation of intracellular calcium pools.
- Subcellularly, HAX1 localizes to multiple compartments including the mitochondrial matrix, endoplasmic reticulum, cell cortex, cytoplasmic vesicles, P-bodies, and the nucleus membrane.
- The protein is ubiquitously expressed and found to be upregulated in oral cancers.
- Post-translational modifications include acetylation and phosphorylation, and alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human HAX1 protein (amino acids 1-279), which may enable detection of endogenous HAX1 across multiple isoforms.
- For Western blot (WB), the predicted band is ~32kDa; verify specificity in positive (e.g., HeLa, HEK293) and negative control lysates.
- In immunofluorescence (IF/ICC), consider the diverse subcellular localizations (mitochondria, ER, cell cortex, nucleus) and use appropriate counterstains to confirm.
- For ELISA, pair with a suitable capture antibody or antigen to assess binding activity; standard curves may be required for quantification.
- Cross-reactivity with mouse and rat is reported, but sequence homology should be validated if working with non-human samples.
