Biological Background: Hyaluronan Synthase 1 (HAS1) Function and Localization
- Hyaluronan synthase 1 (HAS1), encoded by the HAS1 gene, is a glycosyltransferase that catalyzes the synthesis of hyaluronan (hyaluronic acid), a major component of the extracellular matrix.
- HAS1 adds alternating GlcNAc and GlcUA monosaccharides to the growing hyaluronan polymer, playing a critical role in tissue architecture, cell adhesion, migration, and differentiation.
- It is one of three hyaluronan synthase isozymes (HAS1, HAS2, HAS3) in mammals, each with distinct kinetic properties and regulation.
- HAS1 is a multi-pass transmembrane enzyme located in the plasma membrane.
- The protein is widely expressed, with highest levels in ovary, spleen, thymus, prostate, testes, and large intestine; weaker expression in small intestine.
- In addition to hyaluronan synthesis, HAS1 can catalyze the synthesis of chito-oligosaccharides under certain substrate conditions (By similarity).
- The predicted molecular weight is approximately 64.7 kDa (UniProt), often detected at ~65 kDa in western blotting.
