Biological Background: HAPLN1 Function and Localization

  • HAPLN1 (hyaluronan and proteoglycan link protein 1), also known as cartilage-linking protein 1 or proteoglycan link protein, stabilizes aggregates of proteoglycan monomers with hyaluronic acid in the extracellular cartilage matrix.
  • It is a member of the hyaluronan and proteoglycan link protein family, which contains an immunoglobulin-like domain and a hyaluronic acid-binding region.
  • HAPLN1 is secreted into the extracellular space and localized within the extracellular matrix, particularly in cartilage.
  • The protein is widely expressed in various tissues, but weakly expressed in the brain, consistent with its role in cartilage and other connective tissues.
  • Post-translational modifications include disulfide bond formation and N-linked glycosylation, as indicated by its glycoprotein status.
  • Structurally, it consists of an immunoglobulin-like domain and tandem repeats that mediate interactions with hyaluronic acid and proteoglycans, facilitating the formation of stable supramolecular complexes in the ECM.

Experimental Guidance and Technical Tips

  • The immunogen used to raise this antibody is a synthetic peptide spanning amino acids 250-354 of human HAPLN1, which targets an internal region. Consider this when designing experimental protocols, as the antibody may recognize both native and denatured protein.
  • For Western blot (WB) analysis, HAPLN1 has a predicted molecular weight of approximately 40 kDa. We recommend using reducing conditions and validating with appropriate positive controls, such as cartilage tissue lysates or chondrocyte cell lines.
  • The antibody is also suitable for ELISA. For sandwich ELISA development, pairing with another antibody targeting a non-overlapping epitope could improve sensitivity.
  • Given HAPLN1's role in extracellular matrix stabilization, consider evaluating expression in samples from cartilage, connective tissues, or disease models involving osteoarthritis or rheumatoid arthritis.
