Biological Background: HAP1 Function and Localization
- HAP1 (Huntingtin-associated protein 1), also known as Neuroan 1, is a neuronal protein that specifically associates with huntingtin (HTT); the interaction is enhanced by expanded polyglutamine repeats in HTT, linking it to Huntington’s disease pathology.
- It plays a key role in intracellular trafficking, proposed to link HTT to motor proteins and transport cargos, and is involved in vesicular transport within neurons and axons, including early-to-late endosome trafficking and neurite outgrowth promotion.
- HAP1 participates in axonal transport and activity-dependent secretion of BDNF (brain-derived neurotrophic factor) via a complex involving HTT, DCTN1 (dynactin), and SORT1; it also facilitates anterograde transport and membrane insertion of APP, potentially reducing amyloid-beta processing.
- It is involved in delivery of GABA(A) receptors to synapses, dependent on kinesin KIF5, and disruption by mutant HTT may contribute to neuronal dysfunction.
- The protein regulates autophagosome motility by promoting efficient retrograde axonal transport, and is implicated in the regulation of membrane receptor recycling and degradation (e.g., EGFR, androgen receptor, IP3 receptor).
- HAP1 has been suggested to control feeding behavior (via hypothalamic GABA(A) receptors), cerebellar/brainstem development (with AHI1 and NTRK1/TrkA), postnatal neurogenesis (via hypothalamic NTRK2/TrkB), and ITPR1-mediated Ca2+ release.
- It contributes to cytoplasmic retention of REST in neurons through association with DCTN1 and HTT, and may be involved in ciliogenesis and regulation of exocytosis.
- Subcellularly, HAP1 localizes broadly: cytoplasm, axons, presynapse, dendritic spines, dendrites, lysosomes, endoplasmic reticulum, mitochondria, nucleus, autophagosomes, early endosomes, growth cones, neuron projections, secretory vesicles, synaptic vesicles. It is predominantly expressed in brain and selectively in neurons.
