Biological Background: HAO1/GOX Function and Localization
- HAO1 encodes 2-hydroxyacid oxidase 1, a peroxisomal flavoprotein also known as glycolate oxidase (GOX) and glyoxylate oxidase.
- It catalyzes the oxidation of glycolate to glyoxylate using O2 as electron acceptor, producing H2O2; this reaction is central to hepatic glyoxylate detoxification and glycine synthesis. Related references: PMID:10777549 PMID:10978532 PMID:17669354
- The enzyme exhibits broad substrate specificity, also oxidizing glyoxylate and long-chain 2-hydroxyacids (e.g., 2-hydroxyhexadecanoate, 2-hydroxyoctanoate) with lower efficiency. Related references: PMID:10777549 PMID:17669354 PMID:18215067
- It does not utilize L-lactate or 2-hydroxybutanoate as substrates. Related references: PMID:10777549
- HAO1 is localized to the peroxisome matrix, consistent with its role in glyoxylate metabolism.
- The protein is highly expressed in the liver, where glyoxylate detoxification primarily occurs.
- Post-translational modifications include phosphorylation, and it contains an FMN cofactor, classifying it as a flavoprotein oxidoreductase.
- Its predicted molecular weight is ~41 kDa, matching the observed band in Western blotting.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human HAO1; therefore, the antibody is expected to detect full-length HAO1 in denaturing Western blots as a ~41 kDa band.
- For IHC-P, antigen retrieval may be required; test with appropriate positive controls (e.g., human liver tissue) where HAO1 is highly expressed.
- In ELISA, the antibody can be used for detection of recombinant or native HAO1; consider pairing with a capture antibody in a sandwich format.
- Validate cross-reactivity in mouse samples, as indicated; ensure optimal dilutions and conditions are determined experimentally.
