Biological Background: Histone H3.1 Function and Localization
Histone H3.1 is a core component of the nucleosome, which wraps and compacts DNA into chromatin, thereby regulating DNA accessibility for transcription, replication, repair, and chromosomal stability. Nucleosomes consist of an octamer of two copies each of histones H2A, H2B, H3, and H4.
Histone H3 is subject to extensive post-translational modifications (PTMs) on its N-terminal tail, including acetylation, methylation, phosphorylation, and others, collectively forming a “histone code” that governs chromatin dynamics and gene expression.
A recently discovered modification is the serotonylation of glutamine 5 (Q5) of histone H3 (often annotated as 5-glutamyl serotonin). This covalent attachment of serotonin is mediated by transglutaminases and adds a novel layer to the epigenetic landscape.

Histone H3.1 is primarily localized in the nucleus and associated with chromosomes. It is one of the replication-dependent histone variants, expressed during S-phase of the cell cycle.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide immunogen corresponding to a serotonylated sequence around glutamine 5 of human histone H3. It is designed to specifically detect the serotonylated form.
- For Western blotting, acid-extracted histones or nuclear extracts are recommended to enrich the target. A band around 15 kDa is expected.
- In immunofluorescence (IF/ICC), proper permeabilization is essential to access nuclear histone proteins. Include appropriate controls such as unmodified peptide competition or histone mutant cells to confirm specificity.
- Cross-reactivity with mouse and rat has been indicated; however, individual validation is advised for non-human sample types.
- The antibody is also compatible with ELISA; consider testing with serotonylated and non-modified peptides to evaluate differential binding.
