Biological Background: Granzyme A Function and Localization

  • Granzyme A, also known as CTL tryptase, fragmentin-1, Hanukkah factor (HF), and encoded by the GZMA gene (CTLA3, HFSP), is an abundant serine protease found in the cytoplasmic granules of cytotoxic T-cells and natural killer (NK) cells.
  • It induces caspase-independent pyroptosis in target cells after being delivered via the immunological synapse, cleaving after Lys or Arg residues. Related references: PMID:12819770, PMID:32299851, PMID:3257574
  • Once inside the target cell, GZMA cleaves gasdermin-B (GSDMB), releasing its pore-forming moiety to trigger pyroptosis and cell death. Related references: PMID:32299851, PMID:34022140, PMID:36157507
  • It also cleaves APEX1 after Lys-31, destroying its oxidative repair activity. Related references: PMID:12524539

  • Subcellularly, granzyme A is localized to cytoplasmic granules and is secreted upon degranulation.
  • As a disulfide-bonded glycoprotein, it is initially produced as a zymogen and is a well-characterized serine protease with available 3D structures.

Experimental Guidance and Technical Tips

  • For Western blotting, a band of approximately 29 kDa is expected; validate detection using positive control lysates from activated cytotoxic T cells or NK cell lines.
  • In immunofluorescence/immunocytochemistry, permeabilization is required to detect granular cytoplasmic GZMA. Co-staining with perforin or CD8 may confirm cytotoxic cell specificity.
  • When performing ELISA, consider using recombinant human GZMA as a standard and verify linearity in the relevant sample matrix.
  • This antibody shows cross-reactivity with mouse and rat GZMA, but sequence alignment of the immunogen region is advised if quantitative interspecies comparisons are needed.
