Biological Background: Granzyme A Function and Localization
- Granzyme A (GZMA), also known as CTL tryptase, fragmentin-1, and Hanukkah factor (HF), is a serine protease abundantly expressed in the cytosolic granules of cytotoxic T-cells and NK-cells.
- Upon delivery into target cells through the immunological synapse, GZMA triggers caspase-independent pyroptosis by cleaving gasdermin-B (GSDMB), releasing its pore-forming moiety to induce target cell lysis (PubMed:32299851, PubMed:34022140, PubMed:36157507).
Related references:
PMID:32299851 PMID:34022140 PMID:36157507 - It cleaves after Lys or Arg residues (PubMed:12819770, PubMed:32299851).
Related references:
PMID:12819770 PMID:32299851 - Additional substrates include the oxidative repair enzyme APEX1, cleaved at Lys-31 (PubMed:12524539), and the nucleosome assembly protein SET, cleaved at Lys-189 to promote DNA degradation (PubMed:11555662, PubMed:12628186, PubMed:16818237).
Related references:
APEX1: PMID:12524539
SET: PMID:11555662 PMID:12628186 PMID:16818237 - GZMA localizes to cytoplasmic granules and is secreted into the immunological synapse upon target cell recognition.
- The protein is a glycoprotein containing disulfide bonds and is initially synthesized as an inactive zymogen that requires proteolytic processing for activation.
Experimental Guidance and Technical Tips
- For western blotting, expect a band at approximately 29 kDa under reducing conditions; validate in relevant human cell lines such as cytotoxic T cells or NK cells.
- When performing immunofluorescence or immunocytochemistry, consider permeabilization to visualize granular cytoplasmic staining.
- For immunoprecipitation and ELISA, optimize antibody dilution in your specific sample matrix.
- Ensure appropriate positive and negative controls (e.g., GZMA-expressing vs. knockout cell lines) to confirm specificity.
