Biological Background: GULP1 Function and Localization
- Encodes PTB domain-containing engulfment adapter protein 1 (GULP1), also known as cell death protein 6 homolog (CED-6), PTB domain adapter protein CED-6, or protein GULP; belongs to the GULP/CED-6 family.
- Acts as an adapter protein required for efficient phagocytosis of apoptotic cells, facilitating engulfment.
- Modulates cellular glycosphingolipid and cholesterol transport, and may participate in the internalization and endosomal trafficking of various LRP1 ligands such as prosaposin (PSAP).
- Increases cellular levels of GTP-bound ARF6, suggesting a role in ARF6-related signaling and membrane trafficking.
- Localizes to the cytoplasm.
- Widely expressed, with detection in macrophages, pancreas, kidney, skeletal muscle, heart, colon, intestine, lung, placenta, and ovary.
- Annotated with keywords including alternative splicing, apoptosis, coiled coil, cytoplasmic, lipid transport, phosphoprotein, and reference proteome.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (amino acids 1-130) of human GULP1; consider whether this targets all known isoforms as the gene undergoes alternative splicing.
- For Western blot (WB), GULP1 has a predicted molecular weight of 34 kDa; validate the observed band size in your sample system, as post-translational modifications may shift migration.
- In immunohistochemistry (IHC-P), optimize antigen retrieval and detection conditions to achieve specific staining in tissues with known GULP1 expression, such as macrophages or pancreas.
- For ELISA, titrate both the coating antigen and antibody concentrations to establish a suitable working range.
- Cross-reactivity with mouse and rat is indicated; verify the antibody's specificity in these species using positive and negative controls.
