Biological Background: GUCY2D Function and Localization
- GUCY2D, also known as retinal guanylyl cyclase 1 (ROS-GC, GC-E), is a membrane-bound enzyme encoded by the GUCY2D gene in humans. Its multiple disease-associated aliases include LCA1, CORD6, and CSNB1I.
- GUCY2D catalyzes the conversion of GTP to cGMP, a key second messenger in photoreceptor cells, replenishing cGMP levels that are depleted during phototransduction in rods and cones. This activity is critical for visual signal transduction (PubMed:15123990, PubMed:21928830, PubMed:26100624). Related references: PMID:15123990 PMID:21928830 PMID:26100624
- Evidence suggests GUCY2D may also be involved in the trafficking of membrane-associated proteins to the photoreceptor outer segment membrane (By similarity).
- The protein localizes primarily to the photoreceptor outer segment membrane and the endoplasmic reticulum membrane, consistent with its role in sensory transduction and protein processing.
- GUCY2D is a glycoprotein containing multiple transmembrane helices, a disulfide bond, and a signal peptide, characteristic of its membrane integration and secretory pathway processing.
- Mutations in GUCY2D are associated with several inherited retinal dystrophies, including Leber congenital amaurosis (LCA1), cone-rod dystrophy (CORD6), and congenital stationary night blindness, underscoring its essential role in vision.
Experimental Guidance and Technical Tips
- Immunogen design: The recombinant fusion protein immunogen corresponds to the N-terminal region (aa 52-350) of human GUCY2D, which may target the extracellular/lumenal domain. This rational design ensures antibody specificity while avoiding highly hydrophobic transmembrane regions.
- Western blot (WB): A band around 120 kDa is expected based on the calculated molecular weight of ~120 kDa. Due to glycosylation and other modifications, actual migration may vary. Use proper controls and validate in retinal or photoreceptor-derived samples.
- ELISA: The antibody is suitable for ELISA. Consider indirect ELISA format and validate using recombinant protein or cell lysates expressing GUCY2D. Cross-reactivity with mouse and rat orthologs can be advantageous for translational studies.
- Sample preparation: For membrane proteins like GUCY2D, ensure adequate solubilization with appropriate detergents and avoid boiling samples to prevent aggregation. Optimize denaturing conditions for your specific experimental setup.
