- Guanylin, also known as guanylate cyclase activator 2A (GUCA2A) or guanylate cyclase-activating protein I (GCAP-I), is a small secreted peptide hormone encoded by the GUCA2A gene in humans.
- It serves as an endogenous activator of intestinal guanylate cyclase, playing a central role in the regulation of fluid and electrolyte homeostasis within the gastrointestinal tract.
- Guanylin binds to the same receptor region as bacterial heat-stable enterotoxins, stimulating cyclic GMP production and triggering downstream signaling cascades.
- The protein is highly expressed in the ileum and colon and is also detectable in plasma, reflecting its local and systemic endocrine functions.
- The mature peptide contains characteristic disulfide bonds that are essential for maintaining its bioactive conformation.
Experimental Guidance and Technical Tips
- The polyclonal antibody was raised against a recombinant protein spanning amino acids 22–115 of human GUCA2A, covering the proguanylin region and the mature peptide. It is expected to detect both precursor and mature forms in relevant assays.
- For Western blotting under reducing conditions, a band near 12 kDa may be observed; however, post-translational processing could yield smaller fragments. Additional optimization may be required for optimal detection.
- In ELISA applications, this antibody can be employed as a capture or detection reagent. We recommend validating the assay in the relevant sample matrix (e.g., intestinal tissue lysates, plasma) and using appropriate controls.
- Cross-reactivity with mouse and rat guanylin has been demonstrated, enabling utility in rodent models for gastrointestinal research.
