Biological Background: GTF3C3 Function and Localization
- GTF3C3 (General transcription factor 3C polypeptide 3) is a ~101 kDa subunit of the RNA polymerase III transcription factor complex, also known as TFIIIC102, TFIIICγ, or TFiiiC2-102.
- It is an integral, tightly associated component of the DNA-binding TFIIIC2 subcomplex, essential for promoter recognition and transcription initiation by RNA polymerase III.
- The protein directly binds to tRNA gene promoters and virus-associated RNA promoters, enabling assembly of the preinitiation complex at these intragenic control regions.
- GTF3C3 is localized to the nucleus, consistent with its role in transcription.
- The protein contains TPR repeat motifs, is subject to acetylation and phosphorylation, and has been linked to intellectual disability when mutated (UniProt keywords: TPR repeat, Acetylation, Phosphoprotein, Disease variant, Intellectual disability).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 160 amino acids of human GTF3C3; this region may provide specificity for the target in WB applications after denaturing electrophoresis.
- For IHC-P, consider testing various antigen retrieval methods and validating staining in a known positive tissue, as nuclear antigens can be sensitive to fixation and processing.
- When performing IF/ICC, include appropriate nuclear counterstaining and confirm expected nuclear localization in the cell line or tissue under study.
- In ELISA setups, optimize coating antigen concentrations and antibody dilutions to achieve the desired sensitivity and avoid non-specific binding.
