- GTF2H3 encodes the 34kDa subunit of the general transcription and DNA repair factor IIH (TFIIH) core complex, also known as Basic transcription factor 2 34kDa subunit (BTF2 p34), TFIIH basal transcription factor complex p34 subunit, and P34.
- As part of TFIIH, GTF2H3 plays a dual role: in transcription initiation, it is essential for promoter opening and promoter escape by RNA polymerase II; in nucleotide excision repair (NER), it contributes to DNA unwinding around lesions, enabling damaged oligonucleotide excision and repair synthesis.
- TFIIH consists of a core complex and a kinase module (CAK); GTF2H3 is a core component critical for the structural integrity and function of the entire TFIIH assembly.
- The protein localizes to the nucleus, consistent with its functions in transcription and DNA damage response.
- GTF2H3 contains a zinc-finger domain, suggesting metal-binding activity, and undergoes alternative splicing, potentially generating functionally distinct isoforms.
