Biological Background: GSTK1 Function and Localization
- Glutathione S-transferase kappa 1 (GSTK1) is a member of the glutathione S-transferase (GST) superfamily, also designated as GST class-kappa or hGSTK1.
- GSTK1 catalyzes the conjugation of reduced glutathione to various hydrophobic and electrophilic compounds, contributing to cellular detoxification. Significant catalytic activity has been observed using the model substrate 1-chloro-2,4-dinitrobenzene (CDNB). Related references: PMID:14709161 PMID:14742434
- Unlike the majority of cytosolic GSTs, GSTK1 localizes to peroxisomes, indicating a specialized role in organelle-specific detoxification and metabolism.
- Protein expression is ubiquitous across tissues, reflecting its fundamental importance in maintaining cellular redox homeostasis.
- Post-translational modifications, including acetylation, have been documented, and alternative splicing generates multiple isoforms, suggesting complex regulatory mechanisms.
- The three-dimensional structure of GSTK1 has been solved, providing insights into its substrate-binding and catalytic properties.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the full-length human GSTK1 protein (amino acids 1–226), which may enable detection of multiple splice isoforms if present.
- For immunoblotting (WB), an expected band around 25 kDa should be observed; loading controls and molecular weight markers are recommended.
- Given the peroxisomal localization, immunofluorescence and immunohistochemistry protocols may require optimized permeabilization steps to achieve clear organellar staining.
- This antibody has been shown to cross-react with mouse and rat GSTK1, but validation in the end-user’s species and tissue of interest is advised.
- While suitable for ELISA, pairing with a matched detection antibody and titration experiments are necessary for quantitative assay development.
