Biological Background: GSK3α Function and Localization
- GSK3A encodes glycogen synthase kinase-3 alpha (GSK-3α), a constitutively active serine/threonine-protein kinase belonging to the glycogen synthase kinase-3 family.
- Acts as a key negative regulator in glucose homeostasis and Wnt signaling by phosphorylating substrates such as glycogen synthase (GYS1/2), β-catenin, APC, and AXIN1, typically requiring primed phosphorylation. Related references: PMID:11749387, PMID:17478001, PMID:19366350
- Contributes to insulin-regulated glycogen synthesis and mediates the development of insulin resistance by regulating activation of transcription factors. Related references: PMID:10868943, PMID:17478001
- In the Wnt pathway, controls the level and transcriptional activity of nuclear β-catenin. Related references: PMID:17229088
- Facilitates amyloid precursor protein (APP) processing, contributing to amyloid plaque formation in Alzheimer disease. Related references: PMID:12761548
- Acts as a regulator of autophagy by mediating phosphorylation of KAT5/TIP60 under starvation, which promotes acetylation of key autophagy regulators such as ULK1 and RUBCNL. Related references: PMID:30704899
- Phosphorylates the mTORC2 component RICTOR at Thr-1695, facilitating FBXW7-mediated ubiquitination and subsequent degradation. Related references: PMID:25897075
- Involved in neurogenesis, neuronal polarity, and apoptotic signaling; regulated by post-translational modifications including acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- This antibody was raised against a recombinant protein corresponding to the C-terminal region (aa 409–483) of human GSK3α; it is predicted to detect the full-length protein (~51 kDa) in WB applications.
- For WB, use appropriate loading controls and validate the target band in your sample system; consider testing at a range of dilutions to optimize signal-to-noise ratio.
- For IF/ICC, fixation and permeabilization conditions may affect antigen accessibility; it is recommended to perform a pilot experiment to determine optimal conditions.
- For ELISA, ensure proper antigen coating and include negative controls to assess specificity.
- Cross-reactivity has been confirmed with human and mouse samples; if using other species, validation is advised due to potential sequence variation.
