Biological Background: GSK3α Function and Localization
- GSK3α is a constitutively active serine/threonine kinase that negatively regulates glycogen synthase (GYS1/GYS2), CTNNB1/beta-catenin, APC, and AXIN1 by phosphorylation, requiring primed substrate phosphorylation. It contributes to insulin regulation of glycogen synthesis in liver. Related references: PMID:11749387, PMID:17478001, PMID:19366350
- May mediate insulin resistance by regulating transcription factors. Related references: PMID:10868943, PMID:17478001
- In Wnt signaling, GSK3α regulates nuclear CTNNB1/beta-catenin levels and transcriptional activity. Related references: PMID:17229088
- Facilitates amyloid precursor protein (APP) processing and the generation of APP-derived amyloid plaques involved in Alzheimer disease. Related references: PMID:12761548
- Acts as a regulator of autophagy by phosphorylating KAT5/TIP60 under starvation, promoting acetylation of ULK1 and RUBCNL/Pacer. Related references: PMID:30704899
- Phosphorylates mTORC2 component RICTOR at Thr-1695, leading to FBXW7-mediated ubiquitination and degradation. Related references: PMID:25897075
- Necessary for neuronal polarity and axon outgrowth.
- Negatively regulates extrinsic apoptosis and may control cell apoptosis via MCL1 phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1-100). For Western blot, the predicted molecular weight is 51 kDa; validate with appropriate positive and negative controls.
- For IP and ELISA, consider using standard protocols with anti-rabbit secondary antibodies.
- Cross-reactivity with human, mouse, rat implies conserved epitopes; verify in your sample system.
